Cyclin D is required for passage of
the restriction point before S-phase
Antibodies to cyclin D were injected
into specific cells and their passage into S-phase was arrested.
Cells were incubated with the thymidin analoug BrdU
(Bromo-deoxyUracil) to study DNA syntesis. Cells passing into S-phase
will incorporate BrdU into the DNA and can be detected by an antibody
to BrdU.
The same cells are visualised: A.
Blue:
hoechst staining to viualise all cell nuclei. B.
Green:
1° antibody anti BrdU ab made in rabbit IgG type 2°
antibodt anti-rabbit IgG conjugated with FITC. C.
Red: The
injected anti-cyclin D antibody made in goat was detected using a
2° antibody anti-goat IgG conjugated with Texas Red.
